Instructor Notes — Chapter 30

Internal. Teaching companion for Chapter 30: Peptides in Dermatology and Cosmetics.


What this chapter is actually for

On its surface it is a consumer-protection chapter about skincare. It is not. It is the book's cleanest demonstration of a single idea that Chapter 4 introduced abstractly:

Mechanism does not predict effect. Delivery does.

Every compound in this chapter has a coherent, correctly described mechanism. Not one of the ratings turns on mechanism. They turn entirely on whether the molecule arrives where its target lives. And the chapter's ✅ — a 150,000-dalton bacterial toxin — is by far the largest molecule discussed, which detonates the intuition that size is the operative variable.

If your students leave with only one sentence, make it: acetyl hexapeptide-8 and botulinum toxin attack the same molecular machinery and are separated by a needle.


The room you are teaching into

Assume several students own an expensive peptide serum. Some will have bought it recently. Some will have recommended it to friends. A few will have a family member who sells skincare.

This changes how the material must be handled. The chapter models the tone deliberately: concede everything true in the claim first, rate the claim at its strongest, and let the ❌ survive the concession. A ❌ that requires ignoring the true part is not worth having, and students can tell.

Practical consequences for the classroom:

  • Never let the discussion become a competition in debunking. The first student who says "so it's all a scam" should be redirected immediately, gently, and with the GHK-Cu literature.
  • Do not ask students to disclose what they own or spend. Some will volunteer. That is different.
  • Say the useful thing out loud early: the downside here is financial, not medical, and if somebody enjoys their routine that is a legitimate reason to keep it. Students relax considerably once they know the chapter is not going to shame them, and they reason better afterward.

Where students actually get stuck

1. They conflate "penetrates the skin" with "reaches the dermis." This is the single most common error and it is worth five explicit minutes. The tape-stripping point in §30.2 — that the stratum corneum is a waiting room being demolished continuously — usually lands. Ask for a show of hands on "was this molecule detected in living tissue?" before and after.

2. They want ❌ to mean "does not work." Chapter 5's rule 2 will need restating here more than anywhere else in Part 5, because the emotional pull is strong. ❌ describes the evidence. Several ❌ compounds may be ⚠️ in a decade.

3. They assume the split GHK-Cu rating is fence-sitting. It is the opposite: it is the only way to carry both facts. Push them to state the two claims separately and notice that the evidence genuinely meets one and not the other.

4. They think a large sample size rescues a bad design. Exercise AA exists for this. The ranking of the four constructed studies is determined by control and blinding; the study with the most participants is the least informative.

5. They read the marketing language as vague rather than legal. Once §30.7 clicks, students often report they cannot stop noticing it — several will bring in packaging unprompted. Encourage it; it is the most durable thing in the chapter.

6. Strong students overcorrect into cynicism about industry funding. Industry funding does not make a study false. It shapes the population of studies you get to read. That distinction is subtle and worth drawing explicitly.


Timing

A single 90-minute session (if that is all you have): §30.2 barrier and the 500-dalton rule (20 min) → §30.5 the five-step obstacle course (15 min) → §30.6 botulinum toxin as counterweight (15 min) → §30.8 the vehicle problem (25 min) → discussion (15 min). Skip §30.3, §30.4, §30.7 as reading.

Two sessions (recommended): Session 1 — mechanism and barrier: §30.1–§30.6, ending on the size/efficacy inversion. Session 2 — evidence and regulation: §30.7–§30.10 plus case study 2, ending on §30.9's hierarchy.

Lab / workshop hour: exercises W–AB (the constructed studies) run extremely well in pairs, roughly eight minutes each with two minutes of report-back.


Board work worth doing live

The obstacle course. Draw §30.5's five steps as a chain, then assign each step a made-up transmission fraction (say 0.1 each) and multiply on the board. The number that appears is usually more persuasive than any argument. Say clearly that the fractions are invented for illustration — the chapter never assigns real ones, because they are unmeasured, and that is the point.

The two-comparison table. Case study 2's constructed illustration works beautifully on a board. Write the "versus untreated" number, get agreement that it is impressive, then reveal the vehicle row. The reaction in the room is the lesson.

The sentence table. Write four cosmetic claims and four drug claims from §30.7 in two columns without labels, and ask students to sort them. They will do it correctly by instinct and be unable to say why, which is the opening for the structure/function rule.


Assessment suggestions

  • Short written (30 min): exercise P or exercise X. Both reward precision and cannot be bluffed.
  • Problem set: C, H, M, Q, W, X, Y, AA, AC (see answers/ch30.md for marking notes).
  • Take-home: any three daggered exercises, with AB (analyze a real cosmetic study) required.
  • Oral / viva: exercise AF. Ask them to actually say it, out loud, to you. Tone is assessable and this is the chapter where it matters.
