Discussion Guide — Chapter 1

1. "Why do you actually want to know about peptides?"

Open with this, before any content. Every student has a reason and it is rarely the one on the syllabus. Listen for: someone in their family started a GLP-1 drug; an injury that hasn't healed; a supplement they take; a clinic that marketed to them. Naming these out loud early makes the rest of the term concrete and makes it much easier to discuss Part III without anyone feeling personally indicted.

2. "The peptide/protein line is at about fifty residues, and there is no chemical reason for it. Does that bother you?"

Listen for: the move from "that's sloppy science" to "that's a useful convention" to — in the best discussions — "and it's exploitable." Push toward the third. Ask who benefits from the ambiguity, in each direction.

3. "Insulin was used clinically for twenty-five years before anyone knew its structure. What does that tell us about the relationship between understanding and usefulness?"

Listen for: students discovering that mechanism and efficacy are independent. Some will conclude mechanism doesn't matter, which is an overcorrection worth catching — mechanism matters enormously for improving a drug, which is why every advance after 1955 required knowing the sequence. The target is: mechanism is neither necessary for a treatment to work nor sufficient to show that it does.

4. "The collagen case ends without a clean answer. Is that a failure of the case study?"

The most valuable discussion in this chapter. Listen for: discomfort, and then for someone articulating that the discomfort is the finding. Watch for students who want to resolve it by declaring collagen useless — ask them which sentence in the case study licenses that, and let them discover that none does. Also watch for the opposite: students who conclude that since nothing is settled, all positions are equal. Both need the same correction, which is that "unresolved" is a specific state with specific contents, not a synonym for "anything goes."

5. "Should a book about peptides refuse to give dosing information?"

Listen for: a genuine argument on both sides. The case against the book's policy is real — information asymmetry harms people who are going to self-administer regardless, and there is a harm-reduction tradition that would disagree with this book's choice. Do not stack the deck. The strongest version of the objection deserves to be voiced, and students who can articulate it understand the tradeoff better than students who simply approve.

6. "You wrote down what you believe about your peptides and how confident you are. Anyone willing to say one out loud?"

Optional and only in a group with enough trust. Listen for: the confidence numbers more than the beliefs. Ask whether the confident ones are the ones they hope are true. Do not push; the exercise works even unspoken, and it will be called back in Chapter 40 regardless.
